microRNA-122抑制胰腺癌细胞增殖、迁移和侵袭MicroRNA-122 inhibits proliferation, migration and invasion of pancreatic cancer cells

摘要(Abstract):
目的明确microRNA-122(miR-122)在胰腺癌中的表达及miR-122对胰腺癌细胞增殖、迁移和侵袭的作用。方法采用RT-qPCR方法检测20例胰腺癌组织、胰腺癌细胞株(ASPC1、PANC1、MP和SW1990)和人源正常胰腺导管上皮细胞株(HPDE)中miR-122的表达量。将miR-122模拟类似物和阴性对照瞬转PANC-1细胞48 h后,MTT法检测细胞增殖能力,伤口愈合实验检测细胞迁移能力,transwell实验检测细胞侵袭能力。结果与癌旁组织比较,胰腺癌组织中miR-122表达量降低(P<0.01)。与人胰腺导管上皮细胞HPDE比较,4种胰腺癌细胞中miR-122表达量均下降(均P<0.01)。在胰腺癌细胞PANC1中过表达miR-122 48 h后可抑制PANC-1细胞增殖(P<0.01);转染24 h后可抑制PANC-1细胞迁移能力(P<0.01)和侵袭能力(P<0.01)。结论 miR-122在胰腺癌细胞中呈现低水平表达,miR-122抑制胰腺癌细胞增殖、迁移及侵袭能力,其在胰腺癌中可能发挥抑癌基因的角色。
